Changes in mRNA expression of colon mucosa genes during natural aging

چکیده

Background: during natural aging, cellular and molecular changes in the colonic mucosa contribute to chronic damage development of various neoplasia, including colorectal cancer (CRC). Aim: assess age peculiarities mRNA expression samples normal from parts colon. Patients Methods: study included 95 patients. Normal biopsies were derived videocolonoscopy different colon, i.e., right colon (n=91), left (n=95), rectum (n=87). In total, 273 examined. The relative level 63 genes potentially involved pathways CRC carcino-genesis was evaluated by reverse transcription polymerase chain reaction. Results: a linear regression model for predicting passport age, data on gene ex-pression, characterized moderate reliability (R2 0.45, DW test 1.54). Age estimation er-rors arise proportionally its increase, which is associated with accumulation epigenetic changes. Maximum accuracy prediction typical rectal samples. first statistically significant division (primary classification node) patients indi-viduals over or under 60 years provides TERT expression. Telomerase activity association HLA-G5 overexpression older patients, whereas hypoexpression simultaneous granulysin (GNLY) are younger than years. A cascade involving decrease telomerase due cell cycle regulator tumor suppressor P14ARF also aged over. Conclusion: our findings support genetic that occur age. specifics carcinogenesis basis assessing risk. KEYWORDS: expression, mRNA, RT-PCR, mucosa, changes, cancer. Genes with aberrant expression in murine preneoplastic intestine show epigenetic and expression changes in normal mucosa of colon cancer patients.

An understanding of early genetic/epigenetic changes in colorectal cancer would aid in diagnosis and prognosis. To identify these changes in human preneoplastic tissue, we first studied our mouse model in which Mthfr⁺/⁻ BALB/c mice fed folate-deficient diets develop intestinal tumors in contrast to Mthfr⁺/⁺ BALB/c mice fed control diets. Transcriptome profiling was performed in normal intestine...

Protein expression changes of HCN1 and HCN2 in hippocampal subregions of gerbils during the normal aging process

Objective(s): Hyperpolarization-activated cyclic nucleotide-gated (HCN) channels play essential roles in various hippocampal functions, including regulation of long-term potentiation, synaptic plasticity, and hippocampal-dependent cognitive process. The objective of this study was to investigate age-related changes in HCN1 and HCN2 protein expressions in gerbil hippoca...

P-88: Assessing Expression Changes of Some Wnt Pathway Genes During Goat Early Embryonic Development

Background: The developmental competency of embryos is affected by several factors, including the developmental pathways and their elements. In mammalian species including goat, fertilized oocyte undergoes several divisions to form a structure called blastocyst. These events depend on the successful control of temporal and spatial expression of genes involved in genome activation. One of the cr...

Altered hepatic mRNA expression of apoptotic genes during dimethylnitrosamine exposure.

The role of TNFalpha in regulating apoptotic signaling was investigated during subacute, low-dose (5.0 mg/kg) dimethylnitrosamine (DMN)-induced hepatotoxicity. In TNFalpha receptor (TNFR) intact (wild-type, WT) mice following 4 and 7 DMN exposures, hepatic transcripts for TNFalpha and TNFR-1 were elevated as compared to vehicle controls. DMN hepatotoxicity in WT and TNFR-1/TNFR-2 double knockou...

Expression of Melanocortin 4 Receptor mRNA in Male Rat Hypothalamus During Chronic Stress

The effects of chronic stress and glucocorticoids receptor antagonist (RU486) on expression of melanocortin 4 receptor (MC4R) mRNA in arcuate nucleus (ARC) of male rats were evaluated. In this study, adult male Sprague Dawley rats were placed into four groups (n=6/group) stress, RU486, stress/RU486, and control groups. In stress group, the rats were restrained, 1 h/day, for 12 days. In RU486 gr...
